"I definitely have to say these are the windiest conditions I have ever played in," said the 2000 and 2001 champion of the 74-minute victory.
"I am feeling excellent but when you come out in these conditions, you just never know what is going to happen."
Having lost just four games in her opening match, tenth seed Williams breezed through the first set in just 30 minutes.
Kirilenko, the world number 44, traded breaks with Williams early in the second set but the American broke again in the seventh and ninth games to seal her place in round three.
Williams will next play 20th seed Daniela Hantuchova, who beat Maria Emilia Salerni of Argentina 6-1 6-0 on Armstrong Court. Hantuchova hit 13 winners winning 50 points to Salerni's 19 in the 52-minute encounter.
