The Spaniard, who is the defending champion at SW19, cruised through the opening set with Broady only able to muster two games.
But the Brit managed to put up a stiffer resistance in the second set as she took it to 5-5 with Muguruza forced to work very hard to get ahead.
The third seed eventually made the breakthrough and closed out the second set 7-5 with Broady unable to find a response.
Muguruza, who is considered the favourite to defend her title by many, completed victory in 88 minutes after breaking Broady's resistance.
She will next take on the winner of Polona Hercog of Slovenia's clash with Belgium's Alison van Uytvanck in the second round.
