Puig, who made the semi-finals at Nottingham, came into the contest in fine form but was blown away in a first set that saw the British number one take all three of her break opportunities.
The second set was brought to a halt by rain with Konta leading 2-1 and the Puerto Rican returned reinvigorated.
However, Konta prevailed in the tie break to secure her first ever win at Wimbledon to progress to round two after one hour and 14 minutes of action.
"It was a tricky first round, she's playing very well on the grass," said Konta afterwards.
"Conditions didn't make it very easy for both of us. There were a lot of obstacles and I was very happy that I was able to stay focused. I do my best to keep myself present and not rush anywhere. I'm very happy with how I'm serving - it's important to get the first strike in on this surface."
