The 26-year-old did not drop a set through her three qualifying matches, previously beating Cori Gauff 6-4, 6-1, and then Ayano Shimizu 6-1, 6-2.
Watson was seeded 11th in the qualifying draw, but had not won a match at the tournament since she was victorious in the junior competition nine years ago.
Swan had also enjoyed an impressive display so far at Flushing Meadows, defeating Japan's Nao Hibino 6-1, 6-4, after fighting through illness to win a three-set epic against Deborah Chiesa 3-6, 6-4, 6-4.
Watson now joins British number one Johanna Konta as the British representatives in the main draw.
