It was the fourth time Bjorkman and Woodbridge had contested a grand slam final and they once again walked off with the silverware after their triumph at Wimbledon earlier this year.
The two hour victory gave Woodbridge his 77th career doubles title, moving him level on the all-time list with John McEnroe and just one short of the record 78 held by Dutchman Tom Okker.
"This is quite amazing," said Woodbridge, who teamed up with Mark Woodforde to form the most successful doubles partnership in history before his compatriot retired in 2000 after 61 titles together. "Jonas and I have played well together.
"Since we teamed up our goal was to win majors and this is our fourth in three years."
After breaking to take the opening set, the 25 year-old Bryan twins lost their way in the second as Bjorkman and Woodbridge stormed through the next six games to level the match at a set each.
Twice during their slump Mike Bryan called for the trainer to receive treatment on his right leg.
Winners at the French Open in June, the Americans steadied themselves in the third set, which appeared headed towards a tiebreak until Bjorkman and Woodbridge broke to go 6-5 up and then held serve for the victory.
Bob Bryan added a U.S. Open title to the family trophy case when he partnered Katarina Srebotnik of Slovenia in the mixed doubles final beating Russia's Lina Krasnoroutskaya and Canada's Daniel Nestor 5-7 7-5 7-6.
