The second seed was in fine form on Arthur Ashe Stadium and didn't drop serve once against a dangerous opponent as he began his quest to land his second title at Flushing Meadows and a third major title this year following victories at Wimbledon and the Rio Olympics.
British No.1 Murray was slick and clinical in a ruthless display against world No.82 Rosol, winning 83 per cent of points behind his first serve en route to a masterclass under the lights in New York.
"I served very well," said Murray, who hit 11 aces and made just 17 unforced errors.
"I don't think I had any break points against me, which was good, I had good variation on the second serve as well.
"It was a tough start to the match and once I got the break up, I started to relax and play well."
Murray, who improved his win-loss record in 2016 to 51-7, will take on Spain's Marcel Granollers in the second round on Thursday.
