"It feels unbelievable," 27-year-old Bob Bryan said. "It was always our goal at the start of the year. To finish No. 1 for the second time is a great achievement."
The Bryans became only the second doubles team in 50 years to reach each Grand Slam final in one year - following Indians Leander Paes and Mahesh Bhupathi in 1999.
The Bryans won the U.S. Open crown and have 26 career titles overall.
The Bryans, vying to win their third straight Masters Cup title, face Leander Paes and Nenad Zimonjic on Thursday in Shanghai for a spot in the semi-finals.
"It's a good feeling to be the No. 1 team in the world," Mike Bryan said. "We had a great season and hope to finish the year strong here in Shanghai."
