The British team came into Sunday's singles matches with a 2-1 lead, having won both of Friday's singles but lost Saturday's doubles match in a five set thriller.
Britain needed just one point from the final two matches to secure victory at the Braehead Arena, and the Australian Open finalist made no mistake against John Isner as he won 7-6 6-3 7-6.
Isner fought hard throughout but Murray's class was all too evident, and the victory prompted joyous scenes of celebration on the court afterwards.
Though Murray had always been expected to win his two singles rubbers, much of the credit for the win will go to James Ward - whose miraculous victory in Friday night's second singles match, where he came from two sets behind to beat Isner 15-13 in the fifth set, set up the win.
"It feels great. This is a team effort and I think everyone in the team played their part," he said. "The doubles was amazing yesterday and James had an incredible win on Friday.
"It's a pleasure to be a part of this team, all of the staff were incredible this week and it's not just this week but so many the ties over the last six or seven years.
"To be in the quarter-finals for the second year running is incredible and a credit to them.
"I think togetherness was key, everyone was pulling in the same direction together. Liam Broady did a really good speech before the tie to get everyone fired up.
"I've always loved playing the Davis Cup. The whole week is so fun and these guys just make it special.
"I want to say a big thank you to the crowd because it was one of the most special atmospheres I have ever played in. It made a big difference."
Britain will now face France in the quarter-finals, while will be played in July - most likely on grass courts, since Britain have home advantage.
Holders Switzerland, without Roger Federer and Stanislas Wawrinkawent down 3-2 against Belgium in Liege, with David Goffin easing past Adrien Bossel in the critical singles rubber on Sunday night, winning 6-4 6-0 6-4.
Italy, who beat Britain in the quarter-finals last year, went down to Kazakhstan in Astana, where Aleksandr Nedovyesov, ranked 130th in the world, stunned world number 22 Fabio Fognini in five sets to seal a 3-2 victory.
Kazakhstan will travel to former Davis Cup heavyweights Australia who enjoyed a 3-1 victory over 2012 and 2013 winners Czech Republic in Ostrava -- Bernard Tomic clinching the deal with an impressive 7-6(4) 6-3 7-6(5) win over Lukas Rosol on Sunday.
Australia have reached the last eight for the first time since 2006.
World number one Novak Djokovic's Serbia crushed Croatia 5-0 and will take on the winners of the match between Argentina and Brazil in Buenos Aires.
