Istomin, 31, required a medical timeout at the end of the opening set and Edmund took an early advantage, breaking in the very first game of the second set.
Another trusty forehand blow brought up a second break of serve for Edmund as he added another 6-2 set to put himself in firm control against the world number 60.
With Istomin struggling to make any sort of inroads on the Edmund serve, another early break in the third gave the Brit an advantage he enthusiastically pressed home.
Edmund reaches the third round at Melbourne Park for the first time and faces Georgia’s Nikoloz Basilashvili in an eminently winnable encounter.
