Vos (pictured), capped 33 times for the Springboks, retires at the end of the season and will play his last game for Quins in their Premiership clash with Sale at the Stoop.
The former Springbok skipper joined Quins in 2002 and captained the side to the European Challenge Cup title in 2004 and when they won promotion to the Premiership in 2006.
According to the club, former All Blacks fly-half Mehrtens has yet to make a decision on his future. Injuries have restricted him to just eight league appearances this season.
"It's been a fantastic five years of my life," said Vos, who will be making his 139th appearance for the club against Sale.
"I know some people say I retired from the international game at a relatively young age, but it's a decision I've never regretted as I've been made to feel completely welcome at Quins from the day I arrived right through to now.
"Saturday is going to be an emotional day as playing rugby is all I've done for so many years and I'm going to be saying goodbye to some great people.
"However, I'm excited about the future for Quins as we've got a great deal of young talented players coming through, while Dean (Richards) is a very astute man who will develop what we've achieved this season."
Also leaving the Stoop are centre Mel Deane, who is joining Irish province Connacht, prop Peter Cook, hookers James Hayter and Jimmy Richards and back rower Luke Sherriff.
