McGregor will take on Max Holloway at the T-Mobile Arena in Las Vegas, live on TNT Sports and HBO Max.
It will be McGregor's first fight in five years, having last competed in July 2021 when he suffered a leg injury in his trilogy fight with Dustin Poirier.
He has teased a comeback over recent years and was scheduled to face Michael Chandler in 2024, but pulled out two weeks before with a broken toe.
McGregor against Holloway will be a rematch, 13 years after their first bout, when the Irishman won via unanimous decision in Boston.
Before stepping away from the UFC, McGregor had become one of the biggest fighters in the organisation's history, known for his explosive fights and larger-than-life personality.
He became the first fighter in UFC history to hold championships in two divisions simultaneously after knocking out Jose Aldo in 13 seconds at UFC 194 to take the featherweight title before adding the lightweight belt less than a year later with a second-round stoppage of Eddie Alvarez.
McGregor's most memorable moment came at UFC 229 when he fought bitter rival Khabib Nurmagomedov after years of build-up, losing eventually to the Russian.
He did return with a 40-second victory over Donald Cerrone but back-to-back losses to Poirier were his last action in the UFC.
Opposite McGregor will be Holloway who has his own legendary status in the UFC.
He is a featherweight title holder and has a long list of memorable fights over his career, including victories over Aldo, Gaethje, Poirier and Frankie Edgar.
Holloway is ranked No. 4 in the lightweight division so could be in line for a title shot with a victory over McGregor.

Pimblett will fight Frenchman Benoit Saint-Denis at the same event.
The 31-year-old last fought in January this year when he suffered his first UFC defeat at the hands of Justin Gaethje for the interim UFC lightweight title.
Pimblett had been on course to face lightweight champion Ilia Topuria before his loss to Gaethje, with the American now set to face the Spaniard at the White House on June 14. 'UFC Freedom 250' will also be co-headlined by Alex Pereira taking on former interim heavyweight champion Ciryl Gane.
Pimblett, meanwhile, will try to rebuild his title candidacy against Saint-Denis.
The British fighter will face a stiff test against Saint-Denis with the 30-year-old riding a four-win streak and most recently defeating Dan Hooker in February.
Saint-Denis is ranked fifth in the UFC's lightweight standings, while Pimblett sits sixth.
Pimblett and McGregor's bouts will headline the UFC's International Fight Week in Las Vegas.
Elsewhere on the UFC 329 card, unbeaten Luke Riley will take on Kai Kamaka in the featherweight division and fellow Brit Lone'er Kavanagh fights Brandon Royval.
