It is Wales' first defeat in the Nations League, and they were unfortunate to concede a penalty when Ethan Ampadu was harshly penalised for a handball from Viktor Fischer's cross on 62 minutes.
Eriksen slammed home the killer second to condemn the visitors to defeat.
Spurs' playmaker Eriksen had opened the scoring for the home team with a low drive on 32 minutes as the Danes continued their impressive form after reaching the last 16 at the World Cup finals where they lost on penalties to losing finalists Croatia.
Wales had been on a high after defeating Ireland 4-1 in their first game of the tournament on Thursday.
But they were given a technical lesson from Denmark, who were at full strength after several of their senior players had boycotted the 3-0 friendly loss in Slovakia over commercial rights.
