The five players will miss matches against Barcelona in the Champions League and Cardiff in the Premier League.
Eriksen, Vertonghen and Dembele all missed training on Tuesday morning ahead of Wednesday's game against the Liga giants.
As for Alli and Aurier, the pair will be sidelined until after the October international break, according to the club.
There is better news for Spurs with the status of goalkeeper Hugo Lloris, who is available for selection after recovering from a thigh injury.
The Frenchman has been out since August and has an outside chance of being fit enough to play against Barcelona.
With back-up goalkeeper Michel Vorm out with a knee injury, Mauricio Pochettino will have to go with third-choice Paulo Gazzaniga against the La Liga champions if Lloris is deemed unfit to start.
