The 29-year-old Fassa Bortolo rider once again demonstrated his remarkable speed, clinching the 166.7-kilometre stage from Soria to Zaragoza, edging out Spaniard Angel Edo and Julian Dean of New Zealand in a bunch sprint.
It was Petacchi's 12th stage win in a major Tour this year, after amassing six victories in the Giro, four in the Tour de France and now two in the Vuelta.
"I wanted a second win here today because people might say that one win was a fluke, but two is a total nobody can argue with," he said after his victory.
Spaniard Isidro Nozal held on to the overall race lead, with a 50-second advantage over David Etxebarria heading into Thursday's 43.8-km individual time trial.
"Today was a hard day...but tomorrow will be even harder," Nozal said. "I hope I will be able to keep the gold jersey at least as far as the Pyrenees on Friday."
